چکیده

Leptospirosis is an important global veterinary health problem caused by spirochetes belonging to the genus Leptospira. Leptospires are ubiquitous in nature, and therefore able to adapt to both the ambient environment and the renal tubules of chronically infected reservoir hosts. Microscopic agglutination test (MAT) remains a specialized test, which is not generally performed in the routine diagnostic laboratories. In an effort to develop new diagnostic tests to achieve high sensitivity, the focus is mainly on the outer membrane proteins. LipL41 is one among the immunogenic outer membrane proteins that is highly conserved among pathogenic species of Leptopspira. The recombinant based outer membrane protein antigen LipL41 was evaluated for use in the diagnosis of canine leptospirosis by enzyme-linked immunosorbent assay (IgG-ELISA) and latex agglutination test (LAT). The sensitivity and specificity of the developed assays were analyzed with the microscopic agglutination test (MAT) using 221 canine serum samples. The sensitivity and specificity of the ELISA were 83.33% and 93.07%, and the sensitivity and specificity of the latex agglutination test were 95.83% and 96.04% respectively. The assays proved to be as sensitive, specific and accurate as the standard microscopic agglutination test (MAT). key words: enzyme-linked immunosorbent assay, latex agglutination test, leptospirosis, dogs, rLipL41

Evaluation of recombinant Leptospira antigen-based enzyme-linked immunosorbent assays for the serodiagnosis of leptospirosis.

There is an urgent need for development of new serodiagnostic strategies for leptospirosis, an emerging zoonosis with worldwide distribution. We have evaluated the diagnostic utility of five recombinant antigens in enzyme-linked immunosorbent assays (ELISAs) for serodiagnosis of leptospirosis. Sera from 50 healthy residents of a high-incidence region were used to determine cutoff values for 96%...

Evaluation of the recombinant LipL32 in enzyme-linked immunosorbent assay for the serodiagnosis of bovine leptospirosis.

The recombinant leptospiral protein LipL32 was evaluated for use in the diagnosis of bovine leptospirosis by enzyme-linked immunosorbent assay (rLipL32 IgG ELISA). The microscopic agglutination test (MAT) of 150 serum samples from cattle suspected of leptospirosis showed that 125 (83.3%) samples had positive reciprocal agglutination titres, which ranged from 100 to 1600. The highest titres were...

Evaluation of Recombinant Proteins Based Indirect ELISA and Microscopic Agglutination Test for Seroprevalence Study of Feline Leptospirosis

Leptospirosis is worldwide zoonotic disease that is serious problem in south Indian states and now emerging in North India. The focus these days is to develop recombinant outer membrane protein based Enzyme-linked immunosorbent assay (ELISA) for seroprevalence of leptospirosis. The aim of this study was to evaluate indirect ELISA based on recombinant LipL41, Loa22, LipL21, LipL32 and OmpL1 prot...

Application of Recombinant Proteins for Serodiagnosis of Visceral Leishmaniasis in Humans and Dogs

Visceral leishmaniasis (VL) is a zoonotic disease caused by leishmania species. Dogs are considered to be the main reservoir of VL. A number of methods and antigen-based assays are used for the diagnosis of leishmaniasis. However, currently available methods are mainly based on direct examination of tissues for the presence of parasites, which is highly invasive. A variety of serological tests ...
